Hello Jia Qi, First you have to make sure it is a file format that Chimera knows. Then you have to tell Chimera what the format is, for example by how the file is named. The first one is a FASTA sequence file, and Chimera knows that format. If I rename it bpss2276test.fa then it opens fine. The second one is also a sequence file but I can't tell what format it is. It is not correct FASTA format. I don't know where you got it, but maybe you can go there again and save it in FASTA format instead and then tell Chimera what format it is, for example by ending the name with ".fa" Or, could be any other sequence format that Chimera knows, as listed here, with filename according to that format. <https://www.rbvi.ucsf.edu/chimera/docs/UsersGuide/filetypes.html#alignment> I hope this helps, Elaien ----- Elaine C.
